The purpose of this study was to examine the antibacterial potential of the bacterium Pseudoalteromonas piscicida strain B39bio and thereby determine if it can be a source of novel antibiotics. The bacterium has been isolated from Danish coastal waters (Nete Bernbom et al., 2011)) and was grown in Marine Broth and on Marine Agar (Difco 2016(at 25C. Bacterial DNA extracted using: NuclioSpin (740952.250), Maxwell (AS1290) and genomic tip (10223 & 19060). The genome was sequenced using Oxford Nanopore (R9.4.1). The reads from the sequence were of superior quality and gave good coverage. The genome was mined using antiSMASH (antiSMASH 6.0) (Kai Blin, 2021) that searches for gene sequences related to enzymes involved in producing antibiotics and other bioactives. Gene clusters, so-called biosynthetic gene clusters BGCs, likely encoding two secondary metabolites of interest were found in the genome. The BGCs were predicted to encode xenematid and a tolasin variant. By comparing xenemarid with penicillin G theoretically, a shared chemical group (2-phenylacetamid) was found of interest, to have a possible use in semisynthetic antibiotic production. Furthermore, the tolasin variant found was theoretically concluded to be a new variant of an already known antibacterial tolasin molecules, which concludes that is possible to discover novel antibiotic variants using antiSMASH and genome mining.
